William Glover arrived from Virginia at Little River in Perquimans Precinct, North Carolina by 1690. By November of that year he became the clerk of the North Carolina Council. In 1695 he became the clerk of the Perquimans Precinct Court. From 1700 to 1712 he served on the council and also briefly served as acting governor of the colony from 1706 to 1708. He died in North Carolina sometime before October 1712.
